Western blot was performed using Anti-PD-L1 (CD274) Recombinant Rabbit Monoclonal Antibody (JJ08-95) (Product # MA5-32450) and a 40 kDa band corresponding to PD-L1 (CD274) was observed across the cell line and tissue tested, except mouse heart and mouse eye. Whole cell extracts (30 µg lysate) of C2C12 (Lane 1) and tissue extracts (30 µg lysate) of mouse lymph node (Lane 2), mouse heart (Lane 3), mouse spleen (Lane 4), mouse brain (Lane 5), mouse eye (Lane 6), rat lymph node (Lane 7) and rat brain (Lane 8) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556).
